Litle question here, if you decide to kill your heroe to fully barricade a building does it count as a kill in the rule if 4 heroes are killed the zombie win the game.

It certainly does. It doesn't matter how the hero dies -- any time a hero dies, it counts as a kill for the zombie players. This includes sacrifice for barriers, the priest sacrificing himself to cancel a card, blowing yourself or another player up with gasoline or a grenade...
