I was just reading some of the zombie cards on on the flying from wiki & while reading the let's split up card, a scenario came to mind. I just want to know if this could be used.

Since it says Play at the start of a Hero Turn, I'm guessing that it could be played on any of the Heroes, not just the first one. If that's the case, if the zombie player plays it at the start of the fourth hero's turn & there are already two other heroes in the same space or building already, they take a wound.

Does this work or does the card effect only the hero you play it on?

The "Hero Turn" refers to the whole turn, so the card is played before any hero makes a move. So as soon as the Zombie Turn finishes, the card should be played that way it means that the heroes have to do what the card says and split up.
